TGF-β Regulates House Dust Mite-Induced Allergic Airway Inflammation But Not Airway Remodeling

چکیده
“At a Glance Commentary” The current dogma, based largely on indirect evidence, is that TGF-β plays a prominent role in the development of airway remodeling, a recognized feature of chronic allergic asthma. This research contributes to our understanding of the mechanisms regulating inflammation and remodeling in chronic allergic asthma, and suggests that therapeutic strategies to prevent remodeling through TGF-β interference may have detrimental effects.

Transforming growth factor-beta regulates house dust mite-induced allergic airway inflammation but not airway remodeling.
RATIONALE It is now believed that both chronic airway inflammation and remodeling contribute significantly to airway dysfunction and clinical symptoms in allergic asthma. Transforming growth factor (TGF)-beta is a powerful regulator of both the tissue repair and inflammatory responses, and numerous experimental and clinical studies suggest that it may play an integral role in the pathogenesis o...
متن کاملLyn mitigates mouse airway remodeling by downregulating the TGF-β3 isoform in house dust mite models.
Chronic airway remodeling is a serious consequence of asthma, which is caused by complex but largely unknown mechanisms. Despite versatile functions, the role of Lyn in chronic airway remodeling remains undefined. Using Lyn(-/-) mice, we show that continual exposure (for 8 wk) of house dust mite extracts induced a severe phenotype of chronic airway remodeling, including exacerbated mucus produc...
متن کاملAbsence of c-Jun NH2-terminal kinase 1 protects against house dust mite-induced pulmonary remodeling but not airway hyperresponsiveness and inflammation.
Chronic allergic asthma leads to airway remodeling and subepithelial fibrosis via mechanisms not fully understood. Airway remodeling is amplified by profibrotic mediators, such as transforming growth factor-β1 (TGF-β1), which plays a cardinal role in various models of fibrosis. We recently have identified a critical role for c-Jun-NH2-terminal-kinase (JNK) 1 in augmenting the profibrotic effect...
متن کاملSurfactant protein D attenuates sub-epithelial fibrosis in allergic airways disease through TGF-β
BACKGROUND Surfactant protein D (SP-D) can regulate both innate and adaptive immunity. Recently, SP-D has been shown to contribute to the pathogenesis of airway allergic inflammation and bleomycin-induced pulmonary fibrosis. However, in allergic airways disease, the role of SP-D in airway remodeling remains unknown. The objective of this study was to determine the contribution of functional SP-...
متن کاملNeutralization of TSLP Inhibits Airway Remodeling in a Murine Model of Allergic Asthma Induced by Chronic Exposure to House Dust Mite
Chronic allergic asthma is characterized by Th2-typed inflammation, and contributes to airway remodeling and the deterioration of lung function. However, the initiating factor that links airway inflammation to remodeling is unknown. Thymic stromal lymphopoietin (TSLP), an epithelium-derived cytokine, can strongly activate lung dendritic cells (DCs) through the TSLP-TSLPR and OX40L-OX40 signalin...
